چکیده

Damage and degradation of cellular proteins is observed during age-induced seed deterioration, LIsoaspartyl protein methyltransferase (EC 2.1.1.77) is an enzyme hypothesized to play a role in limiting and repairing age-induced damage to proteins. Tomato {Lycopersicon esculentum Mill. 'New Yorker') seeds were assayed for changes in L-isoaspartyl methyltransferase activity during accelerated ageing and after osmotic priming. Accelerated ageing of seeds for 1-4 d at 45 C and 100% relative humidity reduced germination from 94% to 7 1 % , increased the mean time of germination (MTC) from 2.4 to 5.8 d, and was accompanied by a correlative decrease in L-isoaspartyl methyltransferase activity (^ = 0.90). Aged and untreated seeds were primed for 7 d at 20 °C in darkness using aerated solutions of 3% KNO3 or polyethylene glycol 8000 (PEG) with equivalent osmotic potential ( —1.25 MPa). Priming with KNO3 decreased the MTG, but did not improve germination percentage for untreated seeds. Priming did not affect L-isoaspartyl methyltransferase activity in untreated seeds, but restored activity in aged seeds primed in KNO3 to levels near that of untreated seeds. Priming with PEG did not effectively improve the MTG or increase L-isoaspartyl methyltransferase activity. During germination, L-isoaspartyl methyltransferase activity remained constant for 48 h post-imbibition and then declined, suggesting that the enzyme was developmentally regulated and inactivated or degraded as radicle emergence occurred.

Effects of seed pretreatment with 24-brassinolide on physiological and biochemical characters in tomato plants under salt stress

In order to study the diverse effects of seed priming with 24-epibrassinolide (EBL) (0, 0.1, 1 mg/l) to increase tolerance of tomato plants (Lycopersicon esculentum Mill.) to salinity (0, 70 and 140 mM NaCl), the experiments were conducted as factorial based on completely randomized design at greenhouse condition. Results showed that salt stress significantly decreased the growth of tomato plan...

Extended Abstract Introduction: Today, seed priming is widely used to improve seed germination in a wide range of plant species. As a result of seed pre-treatment, several molecular and biochemical changes occur, including increased macromolecule synthesis, enzyme activity and formation of different metabolites. Enzymatic and metabolic activities, synthesis of proteins in quantitative and qual...

Roles of ascorbic acid on physiological, biochemical and molecular system of Lycopersicon esculentum Mill. against salt stress

Tomato is one of the important plants in the world as a food, medicine etc. it is sensitive to abiotic stresses and its crop is affected by them. Moreover, Ascorbic acid (AsA) is one of the universal of plant defence mechanism against the stresses specially salinity and drought to scavenger’s reactive oxygen species. According to global warming and scarcity of water resources, subsequently dryi...

Products Released from Enzymically Active Cell Wall Stimulate Ethylene Production and Ripening in Preclimacteric Tomato (Lycopersicon esculentum Mill.) Fruit.

Enzymically active cell wall from ripe tomato (Lycopersicon esculentum Mill.) fruit pericarp release uronic acids through the action of wall-bound polygalacturonase. The potential involvement of products of wall hydrolysis in the induction of ethylene synthesis during tomato ripening was investigated by vacuum infiltrating preclimacteric (green) fruit with solutions containing pectin fragments ...

Effect of Priming on Germination and Enzyme Activity of Achillea vermicularis Seeds after Naturally and Accelerated Aging

Ageing induces seed deterioration expressed as the loss of seed vigor and/or viability. Priming treatment, which consists in soaking of seeds in a solution of low water potential, has been shown to reinvigorate aged seeds. An experiment was conducted to evaluate the effects of naturally and accelerated aging on seed germination traits and enzyme activities (proxidase, catalase and superoxid dis...
